SENDAI, Japan (AP) -It was one of those games - one of those really big games - that just doesn't sit right for U.S. women's football coach Pia Sundhage.
As far as she is concerned, her team played better than Japan in last year's World Cup final. She still thinks they should have won.
Now, she is looking ahead to the Olympics. And again, Japan is on her mind.
"We have something to prove every time we play Japan," Sundhage said Saturday ahead of a matchup between the two rivals at the Kirin Challenge Cup on Japan's home turf.
The exhibition tournament features three of the top four-ranked teams in the world. The U.S. national team is ranked No. 1 just ahead of Germany, while Japan is No. 3. The other team competing for the title, Brazil, is No. 4.
The U.S. women's squad, which plays Japan on Sunday, goes into the match the favorite. They have a solid record against the Japanese - 22-1-4 - but have lost some big ones lately.
In the victory that made them national heroes, the Japanese team came from behind twice and held on to better the U.S. women in a penalty shootout following a 2-2 draw to win the World Cup final in Germany last summer. It was the team's first win over the United States, and came as the country was still coming to grips with the devastating earthquake and tsunami that ravaged the northern coastline of Japan in March last year.
"In the World Cup, I think we played better than Japan," Sundhage said. "I think we were a little bit unlucky."
But in their most recent game, the Japanese also won 1-0 on March 5 to advance to the final of the Algarve Cup in Portugal. The U.S. had to settle for third place, but went out strongly with a 4-0 win over Sweden. Japan lost the final to Germany.
Both teams are now looking ahead to the Olympics, which start in July in London.
"We are ready and well," Sundhage said. "I don't think we are as well connected as we will be for the Olympics. We are building up for that."
Japan's coach, Norio Sasaki, said his team is at it peak, although it will be playing without star midfielder Homare Sawa, the FIFA women's world player of the year.
"This is a really important chance for us to beat the United States and Brazil ahead of the Olympics," he said. "They are both great teams."
Sundhage said her team will be focused on playing an aggressive game and will try to keep possession of the ball so that they do not have to defend as much against the Japanese, who are a highly skilled, technical squad.
"One thing is absolutely clear - they have always been a technical team, and they are a more physical team today," she said.
After Sunday's opener, the U.S. plays Brazil on Tuesday. The final game, Brazil vs. Japan, is on Thursday.

(NASDAQ:BCOV) is well positioned to benefit from the changing manner of video consumption, evolution of video content on the Internet, proliferation of high speed Internet access, emergence of social media, and an increasing number of different mobile devices.
Brightcove offers Video Cloud, an online video platform that enables its customers to publish and distribute video to Internet-connected devices. In terms of business model, Brightcove prices its products on a subscription basis or as software as a service (SaaS) as it lowers the upfront risk of deploying an in-house, perpetual-based software license solution on top of either new or existing hardware.
The robustness of the product and subscription model has built a highly diversified customer base of over 3,872 in 50 different countries which represents less than 1 percent of potential customers.
"We believe that when corporations weigh the costs and complexities of building a video content distribution platform and the numerous shortcomings of video-sharing Web sites, they are likely to realize that turning to an outside video content distribution platform provider, such as Brightcove, makes the most sense for their video libraries," Breza noted.
The analyst noted that the company has high customer retention as the recurring dollar? rate has been in excess of 93 percent in each quarter of 2011, and trended above 86 percent in each quarter of 2010.
In addition, Brightcove has significant international operations and presence for a company of its size and maturity. During 2011, more than half of all Video Cloud video streams were to international locations. In 2011, the company generated approximately 34 percent of its revenue from customers located outside of North America, up from 32 percent in 2010.
"We expect the current and additional international opportunities to represent a significant area of growth and to trend toward a 50/50% mix over the long term," the analyst said.
Brightcove's total revenue has progressed from $8.5 million in 2007 to $24.5 million, $36.2 million, and $43.7 million in 2008, 2009, and 2010, respectively. Revenue in 2011 totaled approximately $63.6 million, which reflects a year-over-year growth rate of approximately 46 percent.
Breza expects about 21 percent growth in 2012 and believe the company can accelerate its revenue growth rate to approximately 32 percent in 2013.
"Given the early stage of its growth profile, Brightcove has yet to turn a profit, but we estimate that should change over the next several years as the company ramps up and scales against its expense base," said Breza, who has an "outperform" rating on Brightcove shares.

A new study has identified a key mechanism that causes breast cancer to spread. The research, published by Cell Press on March 30th in the journal Molecular Cell, enhances our knowledge about the signals that drive cancer metastasis and identifies new therapeutic targets for a lethal form of invasive breast cancer that is notoriously resistant to treatment.
Amplification of the gene for ErbB2 has been linked with aggressive forms of breast cancer and is associated with a poor outcome. Although ErbB2-positive breast cancers are routinely treated with chemotherapy and the ErbB2-targeted drug trastuzumab (Herceptin), this treatment often fails. "Nearly half of breast cancers with high ErbB2 expression are either initially non-responsive or eventually develop resistance to trastuzumab," explains senior study author, Dr. Tuula Kallunki, from the Danish Cancer Society Research Center. "Therefore, a better understanding of the molecular basis of ErbB2-induced malignancy is critical for further development of targeted treatments."
In the current study, Dr. Kallunki and colleagues discovered that ErbB2 induces expression of proteins called cathepsins. Previous work has linked cathepsins with metastasis of pancreatic cancer, specifically with tumor cell invasion of surrounding tissues and with the establishment of tumor blood supply. Using a three-dimensional model system to study ErbB2-induced cellular invasion, the researchers demonstrated that inhibition of cathepsins prevented ErbB2-driven breast cancer invasion. They went on to identify components of the complex signaling network linking ErbB2 and cathepsin expression with cancer invasiveness.
Taken together, the results demonstrate that the invasive behavior of ErbB2-positive cancer cells is dependent on cathepsin activity and uncover a molecular link between ErbB2 and breast cancer invasion. "Our work has introduced a cellular model system to study ErbB2-induced invasion and identified an invasion-promoting signaling network," concludes Dr. Kallunki. "In addition, our findings provide several potential therapeutic targets against ErbB2-driven invasive cancers."

In London for the premiere of his 3-D rerelease of his Best Picture-winning epic about the doomed ocean liner, which is hitting theaters next month in honor of its 15th anniversary, the famed director talked to E! News about the making of "Titanic," the phenomenon it became and casting stars Leonardo DiCaprio and Kate Winslet.

1. Keep Reshuffling Those Deck Chairs: Cameron, 57, admitted he may have bit off more than he could chew with the 1997 melodrama, particularly given the cost overruns that earned Titanic a flood of negative publicity, so much so that it was being heralded as a bomb before it even opened.
"You know it just seemed like we were doomed," the helmer tells E! News, reflecting back at the long shoot and tortuous postproduction process. "The press were just having their way with us...that we were the biggest idiots in history of Hollywood at least since "Cleopatra"...and so it was difficult. You just had to focus on the work and [hope] that all those rounds just go over the top of the bunker."
2. Capturing Lightning in a Bottle: On "Titanic" becoming a box-office phenomenon -- its $1.8 billion in global ticket sales were the highest-grossing film in history until surpassed by his own film, "Avatar" -- Cameron cited as the reason the fact the flick was able to tap into a set of universal emotions, "speaking to people at a deep kind of universal level that just bypassed language and idiom and culture.
" 'Titanic'...says, 'What would you do if you had an hour or two to live? Would you sacrifice yourself? Would you be that hero? Would you be that guy like Jack or some of those men that put the woman and children onto the lifeboats?'" mused the filmmaker.

3. On Winslet's Audition: Cameron was effusive in his praise for his lead actress. "We cast Kate first and she just blew me away at her screen test," he remembered.
4. Enter the Heartthrob: The King of the World Deep instantly knew the chemistry was right when DiCaprio read for him. "I noticed there was something strange when Leo came in for his first meeting and all of a sudden all the women who worked for me in my entire production company were all in the meeting," Cameron said. "I thought well that's a little strange...and then Leonardo walked in and I went, 'Oh I get it I see what's going on.' Even at that age he was just such a remarkable actor and so accomplished."

5. On Leo's Sudden Fame: Lastly, the artist turned pioneering deep sea explorer revealed that DiCaprio ran away from his iconic role post-"Titanic" because he feared being typecast. "I think it was tough on Leo because he had planned a career as an actor," Cameron said. "I mean, he admired De Niro and that ability to just immerse himself in any character and he knew now...with that iconic status of his character Jack in 'Titanic' that he was going to have to fight against that for a while. So I think he kind of distanced himself from the movie...Now he's fine because he's proven himself."
And so has Cameron both on the screen and off...or at least until "Avatar 2" and "Avatar 3' hit theaters in a few years.

?Considering it may be three, five, even 10 years between purchases of home improvement items?ranging from flooring and gutters to kitchen cabinets and faucets?it?s not so surprising that shoppers turn to new brands when it?s time to replace or upgrade,? says Frank Maher, Integer COO and president of its Midwest office. ?However, home improvement brands can turn shoppers into loyal brand advocates by understanding their shopping behaviors and committing to long-term communication in between those purchases.?
In a report entitled ?Revealing the Motivations that Drive Home Improvement Shoppers,? Integer shares insights around home improvement categories in general, and in-depth findings in three categories: flooring, windows/doors and major appliances. Among its findings are that window/door and flooring shoppers spend the greatest amount of time researching?often by retail site and not brand?before in-store shopping.
The report identifies four shopper types?the Experience Lover, the Fretting Frugal, the Passive Purchase and the Social Adventurer?and reviews how they interact differently with home improvement brands. For instance, the relatively younger Social Adventurer segment is more apt to take action after purchase than other shopper types because of their social shopping behavior.? The research points out the potential these types have to become another medium to advertise a brand.?

CINCINNATI - Researchers have taken a critical step in understanding how allergic reactions occur after identifying a genetic signature for regulation of a key immune hormone, interleukin (IL-13).
Scientists from Cincinnati Children's Hospital Medical Center say the finding opens the potential for new molecular targets to treat allergic disease. They report on March 28 in Mucosal Immunology that a particular microRNA, miR-375, is regulated by IL-13, and in turns regulates how IL-13 induces pro-allergic changes, particularly in epithelial cells in the lung and esophagus.
The data support a role for miR-375 in asthma and in eosinophilic esophagitis (EoE), a severe, often painful food allergy that renders children unable to eat a wide variety of foods. EoE can also cause weight loss, vomiting, heartburn and swallowing difficulties.
"The identification of a microRNA that regulates IL-13-induced changes and inflammatory pathways is a significant advancement for the understanding and future treatment of allergic disease," says Marc E. Rothenberg, MD, senior investigator on the study and director of the Division of Allergy and Immunology and Center for Eosinophilic Disorders at Cincinnati Children's. "MiR-375 is proof of principle that microRNAs are involved in fine-tuning IL-13-mediated responses, which opens up a set of new possibilities for novel therapeutic targets for treatment of allergic disease."
IL-13 induces changes in epithelial gene and protein expression that are important in the onset of many allergic diseases, including EoE. Notably, expression of miR-375 was consistently downregulated after IL-13 stimulated human esophageal squamous and bronchial epithelial cells. Viral overexpression of miR-375 in epithelial cell cultures markedly modified the IL-13-associated immunoinflammatory pathways.
MicroRNAs are short segments of RNA that can regulate whether genetic messengers (mRNAs) are degraded or translated into protein.
In the current study, investigators stimulated esophageal and bronchial human epithelial cells with IL-13 and analyzed for differential microRNA expression. Decreases in miR-375 were observed in the human cells and also in an IL-13 transgenic mouse model. The researchers subsequently assessed miR-375 in patients with EoE, a human allergic disease characterized by IL-13 overproduction, and in healthy individuals.
Interestingly, the researchers found that decreased expression of miR-375 correlates significantly with disease activity, the degree of allergic inflammation and that miR-375 expression normalizes with disease remission. While this suggests miR-375's potential use as a disease activity biomarker for certain allergic diseases, changes in IL-13-mediated inflammatory pathways with viral overexpression of miR-375 in epithelial cell cultures also hint at its therapeutic potential.
Allergic diseases have been on the rise over the past 20 years, with approximately one of every 13 children having food allergies and over 2.5 million children suffering from allergic asthma. Only recently recognized as a distinct condition, the incidence of EoE has also been increasing. Rothenberg and his laboratory team pioneered research showing EoE's reported incidence is estimated to be at least one in 1,000 people. Its hallmark is swelling and inflammation in the esophagus, accompanied by high levels of immune cells called eosinophils.
EoE can affect people of any age, but is more common among young men who have a history of other allergic diseases, such as asthma and eczema. EoE is often first discovered in children with feeding difficulties and failure to thrive, but it is often misunderstood and not well known, delaying proper diagnosis and treatment.

The day after Syrian President Bashar al-Assad agreed to a cease-fire plan proposed by UN envoy Kofi Annan, violence continued in key flashpoint cities, raising concerns that this could become yet another agreement that serves only to buy the regime time.
The Syrian Observatory for Human Rights reported military action in several cities, including Hama and Deraa, where the uprising began more than a year ago, Reuters reports. It also said there was shelling in the city of Homs, which Mr. Assad toured yesterday after his regime's brutal assault of the rebel stronghold last month.
In addition, three Syrian soldiers were killed in clashes with the opposition today in the province of Homs, according to the Associated Press, which cited the same human rights organization. The fighting broke out when the government troops tried to enter the opposition-controlled town of Rastan, it said.
Even before the violence, US Secretary of State Hillary Clinton expressed skepticism that the Assad regime would abide by the cease-fire agreement. "Given al-Assad's history of overpromising and underdelivering, that commitment must now be matched by immediate actions," Mrs. Clinton said, according to CNN. "We will judge Assad's sincerity and seriousness by what he does, not what he says."
Mr. Annan acknowledged that while getting Assad?s agreement was an ?important first step,? implementation would be ?a long and difficult task,? according to Reuters.
His plan has six points: the withdrawal of heavy weapons and troops from populated areas, unimpeded access for humanitarian aid workers, the release of prisoners jailed during the uprising, freedom of movement and access for journalists, a political process between the government and opposition, and allowance of peaceful demonstrations.
Robert Grenier, former director of the CIA counterterrorism center, writes in a commentary for Al Jazeera that Annan?s plan merely helps Assad by buying him time to continue the crackdown.
... does anyone honestly think that the Syrian regime, committed as it is to a programme of violent intimidation and collective punishment, will provide "full humanitarian access", or a daily "humanitarian pause" for those whom it suspects of aiding its adversaries? What are the chances that the tender Mr Assad will release detainees who may promptly rejoin the struggle against him, or that he will permit foreign journalists to freely document his atrocities? Who would want to bet his life, or the lives of those dear to him, that Bashar and his generals will honour a ceasefire, or engage in good faith in a "political dialogue" with those who are challenging their power?
Pursuing such "solutions" is worse than feckless, for it forestalls other, potentially effective actions. By permitting the Syrian regime added time, it is morally equivalent to aiding and abetting Bashar al-Assad.

Such good as can be done in these circumstances will only be done by those who are willing to climb metaphorically into the ring, and to dirty themselves in the process of providing such assistance as is possible to the oppressed of Syria as they struggle to liberate themselves from an unspeakable regime. It will mean taking sides.
The past fourteen or so years have been a great run for flat-panel TV sales, but according to IHS (formerly iSuppli), that run is finally going to turn in the down direction in 2012. Lindsay Lohan is confirmed to make an appearance on "Glee." The announcement comes in a wave of casting news for the hit Fox comedy, which includes a spot for comedian and talk-show host Whoopi Goldberg.
A rep for Lohan confirmed the casting to E! News. In a May episode of the show, Lohan is rumored to be onboard to play herself as a judge at Nationals.
Lindsay's mother, Dina, once took issue with the show for poking fun at her after the show included a scene in which Katie Couric informed Jane Lynch's character, Sue Sylvester, that she beat out Dina and her dog Sparky to make the list of the "world's biggest losers." Dina had said, "It's a shame as the dance numbers are amazing, but the writers need to be nice and more creative as opposed to being hurtful."
Lohan has recently been making strides to get back into the acting spotlight. She hosted "Saturday Night Live" this season and is in talks to play the late Elizabeth Taylor in a small-screen biopic.
As for Goldberg, she'll be in three episodes in May, playing a New York Academy of the Dramatic Arts professor (Carmen Tibideaux) who travels to Ohio to meet with Kurt and Rachel about possible acceptance into the school.
"I've just heard of them, they're really taking over the States," Morrison said. "We are covering one of their songs on 'Glee': 'What Makes You Beautiful.' I need to YouTube it to practice."
A rep for the band hadn't responded to MTV News' request for confirmation by press time.
